The Bhowmick lab is focused on understanding how the tumor microenvironment influences cancer initiation, metastatic progression, and therapeutic resistance. The differences in the microenvironment of the primary and metastatic sites dictate differential therapeutic responses. The translation of our findings has manifested in ongoing interventional clinical trials in limiting lung cancer persister cell populations as well as restoring androgen sensitivity for prostate cancer patients. Companion biomarker development has included liquid biopsy proteomics, circulating tumor cell transcriptomics, and ctDNA analysis.
Seminars
- Showing how carotuximab, a CD105-neutralizing antibody, blocks fatty acid oxidation in castration-resistant prostate cancer cells, increases oxidative stress and reshapes chromatin to restore sensitivity to androgen receptor signaling inhibitors
- Using epigenetic sequencing of circulating tumor DNA from pre- and post-treatment patient samples, supported by CT imaging, to reveal shared downregulation of androgen response, fatty acid oxidation and oxidative phosphorylation pathways across responders
- Highlighting how the kinetics of epigenetic remodeling, with an acute, extensive phase in poor responders versus stable, evenly distributed changes in strong responders, may distinguish treatment response
